RESUMO

1. The objective of this work was to determine the effect of slaughter age and stunning method on the quality of turkey meat from poultry processing plants. 2. One hundred B.U.T. Premium turkeys were divided into 4 groups of 25 animals according to slaughter age (15 or 17 weeks) and CO2 stunning procedure (G1 stepwise: step1: 30% CO2, 15 s; step 2: 55% CO2, 40 s; step 3: 70% CO2, 45 s; G2: progressive increase of the CO2 concentration at a rate of 0.8% per s for 100 s). The quality of the breast meat was determined in fillets taken at different post-mortem times. 3. There were differences between the stunning groups for several variables (pH: 6.01 ± 0.01 and 5.95 ± 0.02; a*: -1.84 ± 0.05 and -2.21 ± 0.04; b*: 4.99 ± 0.15 and 4.68 ± 0.16; drip loss: 0.85 ± 0.02 and 0.71 ± 0.02 for G1 and G2, respectively), while no significant differences were found for L*, cooking loss and texture analysed with a Warner Bratzler Shear Force cell (WBSF). 4. Slaughter age had a significant effect on all the parameters studied (pH: 6.01 ± 0.01 and 5.95 ± 0.01; a*: -2.21 ± 0.05 and -1.88 ± 0.05, b*: 5.50 ± 0.17 and 4.42 ± 0.15; drip loss: 0.71 ± 0.02 and 0.86 ± 0.02, cooking loss: 12.56 ± 0.22 and 14.69 ± 0.16 for turkeys slaughtered at 15 and 17 w, respectively) except on L* and WBSF. 5. The ageing of the meat affected pH, colour values, drip loss and WBSF, with differing degrees of evolution: mean values of L* (39.36 ± 0.35. 45.77 ± 0.20 and 46.30 ± 0.24, for 20 min, 24 h and 7 d post mortem, respectively) and drip loss (0.75 ± 0.03 and 0.84 ± 0.02 for 24 h and 7 d post mortem, respectively) increased, those of a* (-1.77 ± 0.08, -1.94 ± 0.07 and -2.22 ± 0.05 for 20 min, 24 h and 7 d post mortem, respectively) and WBSF decreased (3.73 ± 0.06 and 2.63 ± 0.04 for 24 h and 7 d post mortem, respectively), those of pH decreased in the first 24 h and remained stable for the next 6 d (6.19 ± 0.02, 5.87 ± 0.01 and 5.88 ± 0.01), and those of b* increased in the first 24 h post-mortem and remained stable for the next 6 d (3.26 ± 0.31, 5.86 ± 0.16 and 5.47 ± 0.08). 6. The results revealed no critical differences between stunning methods, and suggest that animals slaughtered at 15 weeks present higher quality meat than those slaughtered at 17 weeks.

This study assessed the influence of genetic type (Embden-Anser anser, EE; Toulouse-Anser anser, TT and F1 cross, ET) for meat characteristics (carcass, meat quality and fatty acid (FA) profiles), of domestic geese "Anser anser domesticus" raised in dehesa as an alternative, organic feeding system. Carcass and breast muscle weight (p<0.01) were greater for the ET group at the same live weight. None of the groups showed differences in the production of fatty liver with this type of feeding. Higher values were found for maximum Warner-Bratzler shear force (between 7.62 and 8.87 kg/cm(2)), which implies the improvement of this parameter. High levels of oleic FAs were obtained, especially for the TT group. The polyunsaturated/saturated FA ratio was highest for the ET group (p<0.001), reflecting the optimum nutritional values as a component of a healthy consumer diet.

A total of 100 young bulls were allotted a 2 × 2 factorial arrangement of treatments to determine the effect of the feeding system (concentrate and wheat straw: T; total mixed ration comprised of the same concentrate, maize silage and wheat straw: TMR) and breed (Limousine: LI; Retinta: RE) on growth performance, carcass characteristics and meat quality. The diets were administrated ad libitum for 193 days. The average daily weight gain was similar (P > 0.05) for both diets, while the LI bulls grew significantly (P < 0.05) more than RE. T bulls showed higher L*, a* and rib bone percentage. TMR bulls showed higher carcass yield, conformation and fatness, and greater changes in ultrasound measurements, except Δ UGMD and rib fat percentage. Instrumental meat quality, except shear force at 1 and 21 days of ageing, was not affected (P > 0.05) by the diets. Breed significantly affected most of the analyzed characteristics.

One-hundred-and-twenty-four young bulls of three Continental beef cattle breeds were used to study the effect of pre-slaughter stress by unfamiliar sounds on carcass traits and quality characteristics of beef aged for three different periods. Stress due to unfamiliar noises had a moderately negative effect on carcass and meat quality. Carcasses from group Stressed had higher (P<0.05) values of pH0, pH24, a*, and Warner-Bratzler shear force, and lower (P<0.05) values of L*, b* and pressure losses than from Unstressed animals. There were significant differences (P<0.05) among breeds in L*, a* and pressure losses. The ageing time had a significant effect on chromatic variables, WBSF and pressure losses. The cooking losses were not significantly affected by any of the three factors discussed. These results emphasize the importance of implementing appropriate management practices during pre-slaughter handling of cattle in order to reduce any possible risk factor for stress, as well as the different sensitivity of the breeds to similar stimuli.

The growth and breed-related changes of rib and rump characteristics in lean beef cattle and the ability of ultrasound to predict carcass traits were investigated. Three hundred bulls from three breeds were scanned monthly (6-7 times) using real-time ultrasound with final scans taken <7 days prior to harvest. The rib and rump ultrasound measurements, except intramuscular fat content, increased (P<0.05) with live weight. Breed affected most of traits. The 12th rib ultrasound measurements showed a weak to high positive correlation (0.131 to 0.976, P>0.05 to P<0.001) with 12th rib measurements in the carcass. Regression equations developed with the ultrasound measurements, explained 97% of the variation in longissimus muscle area, 88% of the variation in fat thickness and 57% of the variation in intramuscular fat content. When last ultrasound scan measurements were excluded from prediction equations, the R(2) significantly decreased. Ultrasound measures "in vivo" are viable options for assessing carcass attributes of lean cattle.

The purpose of this study was to characterize and compare the carcass characteristics, cholesterol concentration, fatty acid composition of intramuscular fat and subcutaneous fat, and meat quality of Criollo Argentino and Braford steers reared in an extensive system, without supplementation, and slaughtered at approximately 400kg live weight. The Braford steers had greater (P<0.05) carcass weight, yield, conformation score, marbling degree, fat thickness and fatness score than Criollo Argentino steers. The tissue composition of the 10th rib was: 68.1% vs. 63.6% muscle, 23.9% vs. 20.4% bone and 8.2% vs. 16.3% fat for the Criollo Argentino and Braford breeds, respectively. The meat of Longissimus muscle from Braford steers was lighter, redder, yellower and more tender than that from Criollo Argentino steers. The meat of Longissimus muscle from Braford steers had a higher fat content, similar protein and ash contents and a lower (P⩽0.001) cholesterol concentration than that from Criollo Argentino steers. The subcutaneous depot was the most saturated, while the intramuscular fat had the most polyunsaturated fatty acids. Intramuscular fat showed the highest ∑h fatty acids, and PUFA/SFA and n-6/n-3 ratios and for MUFA/SFA, 16:0/18:0 and h/H ratios were not significantly different between adipose tissue depots. The influence of breed on the fatty acid profile varies among adipose tissues. In general, both intramuscular fat and subcutaneous fat from Criollo steers contained more unsaturated fatty acids and less saturated fatty acids, than did fat from Braford steers.

Procalcitonin (PCT) is one of the precursors in the synthesis of calcitonin in thyroidal C-cells and other neuroendocrine cells. PCT, among other calcitonin precursors, is elevated in the serum of many conditions associated with a systemic inflammatory response syndrome, even in the absence of the thyroid gland. The aim of our study was to identify PCT-producing extrathyroidal tissues in primate sepsis. In order to induce PCT production, we treated four olive baboons ( papio cynocephalus anubis) with the endotoxin lipopolysaccharide (LPS) from s. typhimurium. We found an increase in serum PCT 3 to 5 hours after LPS injection to levels of 0.2 ng/ml, attaining a peak above 4 ng/ml PCT at 10 hours. In contrast, the untreated baboon had no detectable circulating PCT in the serum. In one animal, additional LPS boosting after 24 hours did not increase serum PCT further. Soluble proteins were extracted from different organs, fractionated by C18 extraction, and PCT was measured in an immunoluminometric assay (ILMA), which was specifically developed for this study. PCT concentrations above 0.2 ng/g of wet tissue were found in a variety of organs in LPS treated baboons, but not in the control baboon. Organs and tissues with the highest PCT concentration included liver, kidney, aorta, fat, ovaries, bladder and adrenal gland. RT-PCR confirmed an extrathyroidal origin of PCT. Importantly, CT-mRNA expression was found in liver, lung, kidney, adrenal, colon, skin, spleen, brain and pancreas. In conclusion, our data confirm previous findings in hamsters, indicating an extrathyroidal origin for PCT in sepsis. Our primate model offers a valuable tool for further investigation of PCT's pathophysiological role and its immunoneutralization as a therapy for sepsis.

BACKGROUND: Recently, we reported an unexpected ubiquitous expression of calcitonin (CT)-mRNA in a hamster peritonitis model of sepsis. Using this animal model,we undertook a study to further investigate the pattern of expression of the calcitonin I (CALC-I) gene and CT gene-related peptide (CGRP)-mRNA in sepsis. METHODS: Live Escherichia coli impregnated in agar pellets were implanted in the peritoneal cavities of hamsters. Twelve hours after sepsis induction, the septic and healthy control animals were sacrificed and tissues and peritoneal macrophages were collected. CGRP-mRNA content was evaluated by reverse transcription polymerase chain reaction (RT-PCR), quantitated by the Taq-Man technique, and compared with the mRNA expression of CT, tumor necrosis factor alpha (TNF-alpha), and interleukin-6 (IL-6). The 5' untranslated regions of the mRNA and potential alternative splicing sites were identified by 5' rapid amplification of cDNA ends. RESULTS: We found a tissue-wide, ubiquitous and uniform expression of CGRP-mRNA in all septic tissues examined. CGRP-mRNA was detectable by RT-PCR in various extraneuronal and extrathyroidal septic tissues, but not in healthy control tissues. As found for CT-mRNA in our earlier studies, CGRP-mRNA seemed to be more specifically up-regulated as compared with other classical cytokines (ie, II-6 and TNF-alpha). Importantly, the 5' untranslated sequence in control and septic thyroid was similar to the sequence obtained from septic spleen. CONCLUSIONS: We postulate the presence of microbial infection-specific response elements in the CALC-I gene promotor, which, upon a specific stimulus, override the tissue-selective expression pattern. This new form of endocrine plasticity may be of importance in the response to systemic inflammation.

RESUMO

The authors report a case of thoracic spinal cord stab injury with neurologic impairment that was treated surgically after injury. A literature review and case analysis indicate that surgical extraction of foreign bodies retained within the spinal canal is indicated to avoid infection, delayed myelopathy, and neurologic loss. The amount of motor and functional recovery for incomplete injuries after spinal cord stab wound can be strikingly good despite pathologic changes to severely damaged areas, and removal of retained intraspinal metallic fragment can improve this neurologic outcome. Open removal of the knife seems preferable to avoid bleeding and infection.

PIP: In order to study the cyclic variations in the immunoglobulins of the human vaginal fluid, vaginal secretions were obtained from 12 volunteers during the follicular phase (Days 7-12 of cycle) and from 12 during the luteal phase (Days 18-25 of cycle). At follicular phase, immunoglobulin G was present in all samples, and immunoglobulin A similar to serum immunoglobulin A was found occasionally. At luteal phase, immunoglobulin G apparently occurred at a greater concentration, and another immunoglobulin A, different from the serum type, was found. No immunoglobulin M was detected in any sample. It is postulated that the cyclic variations in immunoglobulins of human vaginal fluid, which occur under normal conditions could be 1) related to the plasma cells found in the vaginal mucosa and 2) mediated by the ovarian hormones.^ieng
